When a rocket intended to go to Mars blasts off by accident with Lester and Orville on board, it ends up in New Orleans on Mardi Gras where they mistake the people in costumes for aliens, whereupon they run into two criminals on the run who force them to take them to outer space, landing on Venus, which is inhabited only by females.
